Introduction: RF-amide–related peptide-3 (RFRP3), in addition to its inhibitory role in reproductive function, regulates appetite and energy metabolism. This study investigated the effects of a high-fat diet (HFD) and high-intensity interval training (HIIT) under HFD conditions on serum and hypothalamic RFRP3 levels in laboratory mice. Materials and Methods: In this experimental study, we randomly assigned 18 male C57BL/6 mice to three groups: healthy control, high-fat diet (HFD), and high-fat diet plus high-intensity interval training (HFD+HIIT), with six animals in each group. The HFD group received a high-fat diet for 20 weeks. The HFD+HIIT group received the same diet and, during the final eight weeks, performed high-intensity interval training on a treadmill. At the end of the intervention, RFRP3 levels in serum and hypothalamic tissue were measured using enzyme-linked immunosorbent assay (ELISA). Results: High-fat diet, both alone and in combination with HIIT, resulted in a significant reduction in serum and hypothalamic RFRP-3 levels compared to the control group. However, direct comparison between the HFD+HIIT and HFD groups showed that HIIT did not significantly change serum or hypothalamic RFRP3 levels (serum: p=0.247; hypothalamus: p=0.691). Conclusion: Although the high-fat diet was associated with reduced serum and hypothalamic RFRP3 levels, the HIIT protocol used in this study did not attenuate these changes, suggesting that while RFRP-3 is sensitive to high-fat diet intake, the HIIT intervention did not significantly modulate it. |
